Update (5:25 p.m.): Police say both drivers were taken to local hospital.
The female driver of the car was later airlifted to a Toronto hospital with "serious" injuries, according to Barrie police communications co-ordinator Peter Leon.
The two drivers were the only occupants in their vehicles, he added.
The on-scene investigation continues.
More information is expected to be released Wednesday morning.

Members of the Barrie police traffic unit are on the scene of a "serious" crash on Ellis Drive in the city's south end this afternoon.
Around 4 p.m. Tuesday, police said the collision involves a car and a transport truck.
Ellis Drive is closed at Welham Road for the investigation.
Police say Ellis Drive will be closed until further notice to allow the investigation to safely take place.
